Neurons that have been derived from various types of stem cells have recently undergone significant study due to their potential for use in various aspects of biomedicine. In particular, glutamatergic neurons differentiated from embryonic stem cells (ESCs) potentially have many applications in both basic research and regenerative medicine. This review summarized the literatures published thus far and focused on two areas related to these applications. Firstly, these neurons can be used to investigate neuronal signal transduction during differentiation and this means that the genes/proteins/markers involved in this process can be identified. In this way, the dynamic spatial and temporal changes associated with neuronal morphology can be investigated relatively easily. Such an in vitro system can also be used to study how neurons during neurogenesis integrate into normal tissue. At the same time, the integration, regulation and functions of extracellular matrix secretion, various molecular interactions, various ion channels, the neuronal microenvironment, etc., can be easily traced. Secondly, the disease-related aspects of ESC-derived glutamatergic neurons can also be studied and then applied therapeutically. In the future, greater efforts are needed to explore how ESC-differentiated glutamatergic neurons can be used as a neuronal model for the study of Alzheimer’s disease (AD) mechanistically, to identify possible therapeutic strategies for treating AD, including tissue replacement, and to screen for drugs that can be used to treat AD patients. With all of the modern technology that is available, translational medicine should begin to benefit patients soon.  相似文献

There has been an increasing prevalence of neurodegenerative diseases with the rapid increase in aging societies worldwide. Biomarkers that can be used to detect pathological changes before the development of severe neuronal loss and consequently facilitate early intervention with disease-modifying therapeutic modalities are therefore urgently needed. Diffusion magnetic resonance imaging (MRI) is a promising tool that can be used to infer microstructural characteristics of the brain, such as microstructural integrity and complexity, as well as axonal density, order, and myelination, through the utilization of water molecules that are diffused within the tissue, with displacement at the micron scale. Diffusion tensor imaging is the most commonly used diffusion MRI technique to assess the pathophysiology of neurodegenerative diseases. However, diffusion tensor imaging has several limitations, and new technologies, including neurite orientation dispersion and density imaging, diffusion kurtosis imaging, and free-water imaging, have been recently developed as approaches to overcome these constraints. This review provides an overview of these technologies and their potential as biomarkers for the early diagnosis and disease progression of major neurodegenerative diseases.  相似文献

北海道では1980年代から2010年代まで,水稲圃場栽培期間である5–9月の気温は年代とともに上昇した。そこで,直近の2010年代(2010—2019年平均)と比べて, 2つの2030年代の予測気象から,既報の関係式より水稲生育を予測した。その結果,2030年代では2010年代に比べ,限界移植日(移植早限)が水稲栽培17地域の平均で8~9日早い。また,早限出穂期が1~5日早く,晩限出穂期が1~5日遅く,安全出穂期間が2~10日長い。出穂期は1~3日早い。出穂期から晩限出穂まで2~9日長いため,遅延型冷害の発生がやや少ない。生育期別気象は,出穂前24日以降30日間では生育が早いため平均気温が同じかやや低い。出穂前10日以降40日間および出穂期以降40日間では平均気温がやや高く,日射量はやや少ない。そのため,玄米収量は96~98%とやや低く,潜在収量性を示す気候登熟量示数は同じである。障害不稔発生に関係する穂ばらみ期冷害危険期の平均気温はわずかに低いかほぼ同じであるため,冷害発生の危険性は残る。一方,不稔発生をもたらす低温域の出現頻度には,地域間で差異がある。精米蛋白質含有率は同じであるが,アミロース含有率はやや低く,やや良食味である。米粒外観品質では被害粒歩合と着色粒歩合は一定の傾向がなく,未熟粒歩合はやや高い。精米白度は同じであるが,玄米白度はやや高い。以上の予測に対する技術的対応方向を示した。  相似文献

The use of agro-industrial waste as substrates for mushroom cultivation is considered a promising management strategy for reducing and valorising these wastes, simultaneously reducing the cost of mushroom cultivation. In this study, oyster mushroom Pleurotus ostreatus were cultivated on twelve substrates composed of either tea waste, lime sawdust, alder sawdust, hornbeam sawdust/shaving, wheat stalk-straw, wheat bran or their composite to determine the proximate composition and accumulation of thirteen elements in their fruiting bodies. The proximate composition of P. ostreatus did not show a significant difference, regardless of the employed substrate. (one-way manova ; F(66, 107) = 1.329, Wilk’s λ = 0.041, P > 0.05). However, their chemical element contents show a statistically significant difference (one-way manova ; F(132, 418) = 32.163, Wilk’s λ = 0.000, P < 0.05). These results were supported by discriminant function and principal component analyses. The highest mean concentrations of six of twelve elements (i.e., Ca, Mg, Na, Zn, Cd and Cr) were recorded in P. ostreatus cultivated on the lime-sawdust substrate. Three health indices viz., estimated daily intake, target hazard quotient (THQ) and total THQ were applied to determine the risk to human health via the consumption of P. ostreatus, suggesting that they are safe for human consumption.  相似文献

Thin film of WO3 has been deposited on conductive fluorine tin oxide substrate using the hydrothermal technique. The film's microstructural, morphological and optical properties have been identified using X-ray diffraction, atomic force microscope, and spectrophotometer. The obtained results have confirmed the nanocrystalline structure of the as-received WO3 thin film with crystallite size ≈63.4 nm. Analysis of the absorption coefficient using Tauc's model shows the possibility of direct and/or indirect allowed transition with energy gaps 3.95 and/or 3.45 eV, respectively. The refractive index has been determined by different methods showing the average value (2.2 and 2.3 corresponding to the direct and indirect transitions, respectively). The nonlinear refractive index and third-order nonlinear optical susceptibility have been determined, showing the high polarizability of WO3/FTO with radiation to be promising for different optical devices and applications.  相似文献

A predictive model based on Fick's second law for radial diffusion is proposed and validated for modeling the diffusion of fragrance materials. A pure component, two binary systems, and a ternary system were used for validation. The model combines the prediction model to represent the liquid phase nonidealities, using the UNIFAC group contribution method, with the Fickian radial diffusion approach. The experimental headspace concentrations were measured in a diffusion chamber using the solid-phase microextraction technique and quantified using gas chromatography with a flame ionization detector. The numerical solutions were obtained along with an analytical model considering constant surface concentration. The odor intensities of the studied systems were calculated using Stevens' power law and the strongest component model, respectively. The numerical simulation presented good adherence to the experimental gas concentration data. The proposed methodology is an efficient and validated tool to assess the radial diffusion of fragrance and volatile systems.  相似文献

Substantial compressive deformation occurs in the gas diffusion layer (GDL) under the pressure applied during the fuel cell assembly. The GDL deformation has a direct impact on the efficiency and performance of the fuel cell since it leads to the alteration of the GDL microstructure and porosity. This makes the accurate characterization of the GDL compressive behavior crucial for analyzing the fuel cell performance and its optimal design. In this paper, analytical, experimental, and numerical methods have been employed to comprehensively study the constitutive law of the GDL under compression. Starting from the recently developed stress-density relations, the constitutive stress-strain equations are derived for the GDL and the relation between the stress-density and stress-strain laws are revealed. Experimental compression tests have been performed on GDL samples and the capability of the proposed constitutive law in capturing the real behavior of the material has been proved. It has been observed that the simplifying assumption of constant zero Poisson's ratio in the through-plane direction made in many previous studies cannot accurately represent the GDL material behavior and a modification is proposed. The developed constitutive law has been successfully implemented in a finite element model of the GDL-bipolar plate assembly in the fuel cell structure and the variations of the GDL porosity, density, and through-plane Young's modulus and Poisson's ratio have been investigated for different vertical displacements of the bipolar plate.  相似文献

For the United States Nuclear Regulatory Commission and the reactor licensees it regulates, there are a number of contemporary issues associated with the back end of the fuel cycle including, the agency's revision to its “Waste Confidence” decision and the path-forward for high-level waste disposal. Additionally, the 2012 Blue Ribbon Commission on America's Nuclear Future recommendations, the future of reprocessing, consolidated interim spent fuel storage, and maintaining technical competence within the NRC in challenging budgetary conditions are addressed. I conclude that there is confidence in the feasibility of safe storage of spent nuclear fuel following the licensed operational life of a reactor and any change in high-level waste policy will require Congressional action to amend the Nuclear Waste Policy Act.  相似文献
